Tokyo, Japan - June 17, 2015 - Sony Corporation ("Sony") today announced the launch of its SA3800 spectral cell analyzer, a new flow cytometer equipped with a newly developed autosampler that features 3-dimensional drive (X, Y, Z axes). The SA3800 combines ease of operation with full automation, and its autosampler delivers high-speed analysis and processing performance.

A flow cytometer is a device that irradiates cells stained with a fluorescent reagent with a laser and analyzes the types and properties of the cells by measuring fluorescence and scattered light. Flow cytometers are widely used in cellular research fields such as immunology, oncology, and regenerative medicine, as well as in drug discovery. Particularly in realms like drug discovery and biomarker1 discovery, which require analysis of many types of cells, there is a real need for the capacity to analyze large numbers of specimen rapidly, efficiently, and accurately.

With conventional autosamplers, the well plate moves horizontally, while the sample suction probe2 moves vertically to collect the samples. The consequence of this is that the sample must traverse a relatively long distance before reaching the measuring unit, and this can cause issues with processing speed and carryover ratio (the rate of cross contamination between samples). The SA3800's newly developed autosampler employs a stationary sample suction probe and a well plate that moves in three dimensions (X, Y, Z axes), thus speeding the sample acquisition and analysis process. Additionally, the probe has a built-in self-washing function that further reduces the carryover ratio to 0.1% or less.3 These capabilities enable the SA3800 to realize stable yet high-speed sample collection, which is a much sought after feature for the analysis of large numbers of samples.
The optical unit employs Sony's original spectral analysis system for detecting fluorescence waveforms, which has a proven track record as a technology installed in Sony's SP6800Z series4 cell analyzer already on the market. The adoption of a highly sensitive 32 channel photomultiplier tube (PMT) makes possible the detection and separation of the previously unmeasurable factor of cell autofluorescence5. It also greatly simplifies the time-consuming process of fluorochrome correction and ensures that results are verifiable, regardless of who is operating the device.

Sony will exhibit and demonstrate the SA3800 at the 30th Congress of the International Society for Advancement of Cytometry (CYTO 2015), to be held from June 26th in Glasgow, UK.

2. A tube for extracting cell samples
3. Measured using Sony's own evaluation method in which beads and cells are measured alternately

5. A weak fluorescence emitted by the cells themselves

Main features of the SA3800

1. Newly developed "3D AutoSampler"
The SA3800's autosampler employs 3-dimensional drive to collect cell samples rapidly and reliably. When used in concert with 96-well or 384-well plates6, automated analysis of large numbers of specimen can be accomplished rapidly and with low carryover.
6. A well plate is a receptacle consisting of a rectangular panel with multiple rows of "wells" for storing a variety of individual samples such as cell solutions. For example, a 96-well plate has 12 rows of 8 wells each.
Main capabilities of the "3D AutoSampler"
- High-speed autosampling function: 25 minutes per 96-well plate
- Low carryover: 0.1% or less
- Sample probe with built-in self-washing function
- Compatible with 96-well plates, 384-well plates, and 5 mL tube racks
- Sample agitation function by shaking; sample cooling function
2. Highly accurate and reliable analysis with Sony's original spectral analysis function
1) Without applying optical filtering, the fluorescence wavelengths are divided by color using Sony's uniquely designed prism. In combination with the 32 channel photomultiplier tube (PMT), this enables highly accurate measurement of fluorescence waveforms.
2) Sony's original analytic algorithm
An analytic algorithm developed by Sony is used to divide multiple fluorochrome signals into discrete color information based on waveforms. This data can then be analyzed according to characteristics such as intensity. The analytic algorithm can distinguish between data from fluorescent reagents with very similar waveforms and very closely located fluorescence wavelength peaks, which was difficult to achieve with conventional optical filtering.
3) Autofluorescence detection
Measurement of large numbers of specimen may be complicated by inconsistent background fluorescence due to factors such as individual samples having different autofluorescence levels. This can make it difficult to compare samples quantitatively. Sony's spectral analysis function addresses this by discerning autofluorescence, identifying it as a color, and separating it out from the other signals. This thus overcomes the issue and contributes to more reliable analysis.
3. Flexible optical system accommodating up to four lasers
As needed, 405 nm, 561 nm, and 638 nm lasers can be added to supplement the standard 488 nm laser (for a total of up to four lasers). It is also possible to upgrade the SA3800 with additional lasers at a later date.
